Fenretinide Acts as Potent Radiosensitizer for Treatment of Rhabdomyosarcoma Cells

Fusion-positive rhabdomyosarcoma (FP-RMS) is a highly aggressive childhood malignancy which mainly treated by conventional chemotherapy, surgery and radiation therapy. Since radiotherapy associated with high burden of late side effects in pediatric patients, addition radiosensitizers would be beneficial. Here, we thought to assess the role fenretinide, potential agent for FP-RMS treatment, as radiosensitizer. Survival human cells was assessed after combination therapy fenretinide ionizing (IR) cell viability clonogenicity assays. Indeed, this found significantly reduce compared single treatments. Mechanistically, accompanied enhanced production reactive oxygen species, initiation cycle arrest induction apoptosis. Interestingly, treatment also triggered new form dynamin-dependent macropinocytosis, previously described fenretinide-only cells. Our data suggest that acts IR induce death therefore might represent novel radiosensitizer disease.
